Cartilage intermediate-layer protein (CILP)
CILP is a monomeric extracellular matrix glycoprotein
expressed mainly in the middle zones of human
articular cartilage and in the meniscus,tendons,ligaments,
synovial membrane,and intervertebral disc (41).
CILP acts as a negative regulator of TGF_,which
controls the expression of aggrecan,type II collagen,
and extracellular matrix proteins important in the mechanism
of fibrosis of the intervertebral disc annulus and in
maintaining nucleus pulposus integrity.Key information
about CILP is reviewed succinctly in an article by Mori
et al (41).Human articular cartilage CILP expression
increases with aging,in the early and late stages of OA,
and in calcium pyrophosphate dihydrate crystal deposition
disease.A functional SNP in CILP is associated
with susceptibility to lumbar disc disease in the Japanese
population (Table 3).This study also showed increased
localization of CILP in degenerated intervertebral disc.
Perhaps the mutant CILP protein affects chondrocyte
homeostasis by exerting an increased inhibitory effect on
TGF_1.These epidemiologic findings were not replicated
in Finnish and Chinese cohorts affected by lumbar
disc disease,which might reflect ethnic differences in the
risk factors for lumbar disc disease (Table 3).
